PURPOSE:In the titled method of measurement using a novel-amylase substrate, to provide method of measurement with a small error and improved reproducibility, by using an alpha- and/or beta-phenylmatooligoside having a modified nonreducing end as a substrate. CONSTITUTION:An alpha- and/or beta-phenylmaltooligoside (formula) obtained by modifying a nonreducing end of maltooligosaccharide such as maltotetraose, maltopentaose, etc. with a halogen atom, -X-R<3> or glucopyranose residue is used as a substrate. A test specimen is brought into contact with it in the presence of alpha-flucosidase and/or glucoamylase, and, if necessary, beta-glucosidase, and a free phenolic compound is measured, to measure alpha-amylase activity in the test specimen.
