PURPOSE:To enable quantitative determination with high sensitivity from a microtrace quantity to a high concn. without the need for dilution and to determine quantitatively the H2O2 formed by an enzyme reaction as well in the stage of determining quantitatively the H2O2 with a triphenyl methane deriv. as a color developing component by adding an azide to a sample to adjust color sensitibity. CONSTITUTION:The triphenyl methane deriv. expressed by the formula [R1, R2, R3, R4 are lower alkyl, X1, X2 are -O(CH2)nSO3M1, M1 is H, alkali metallic ion or NH4<+>, n is 2-4 or one is -O(CH2)nSO3M1 or SO3M2, M2 is the same as M1 and the other is H] is added as a color developing component together with an azide, particulary sodium azide at about 0.01-0.5% into a color developing reagent liquid. Although there is a need for diluting the sample in the stage of using only the triphenyl methane deriv., the measurement with good linearity to 100mg/l with high sensitivity from a microtrace quantity is made possible without the need for such dilution. The exact measurement of the component such as chloesterol in a bio-fluid is made possible by generating the H2O2 by the enzyme reaction and measuring the H2O2. |
