PURPOSE:To obtain a reagent suitable for measuring amylase isozyme, capable of determining separately amylase derived from the pancreas and amylase derived from spittle simply in a short time, by combining two reagents. CONSTITUTION:The first reagent containing alpha-glucosidase, hexokinase, glucose-6- phosphate dehydrogenase, adenosine triphosphate, nicotinamide adenine dinucleotide, magnesium acetate and amylase inhibitor is combined with the second reagent containing maltopentaose, and an amylase activating substance such as sodium chloride, potassium chloride, etc. is added to at least one of both the reagents. The reagents are used as buffers solution which are usually adjusted to 6-8pH.
