PURPOSE:To enable quantitative determination of the concn. of the acralvicine in a blood in short time with good accuracy by using the antibody from an animal immunized by using the specific deriv. of acralvicine and the labeled deriv. in quantitative determination of the acralvicine which is an antitumor antibiotic. CONSTITUTION:The carbonyl group in the synellose A part of the acralvicine expressed by the formula used for the treatment of the acute leukemia, the malignant lymphgranulomatosis, the cancer of the stomach, the lung, the breast, etc. is aminated to synthesize 4'''-deoxo-4'''-aminoacralvicine (abbreviated as acralvicine). Such acralvicine of which the amino group is bound with, for example, bovine serum albumin is injected to a rabbit and the blood is drawn therefrom to obtain an antiserum. On the other hand, the aminoacralvicine which is labeled by beta-D-galactosidase is synthesized to obtain a labeled antigen. After the patient's serum and a specified amt. of the labeled antigen and the antiserum are brought into reaction, an antirabbit IgG goat serum is added thereto and after incubation, the serum is centrifugally separated and a soln. of 7-beta-D galactopyranocyloxy-4-methylcumarine is added thereto as a substrate. The fluorescent intensity of the formed 7-hydroxy-4-methylcumarine is measured. |
