PURPOSE:To obtain NAD(P)H oxidase having high activity at normal temperature and thermal stability, by cultivating Bacillus stearothermophilus of thermophilic bacterium. CONSTITUTION:NAD(P)H Oxidase capable of oxidizing both of reduced nicotinamide adenine dinucletide and reduced nicotinamide adenine dinulceotide phosphate is collected from the culture of Bacillus stearothermophilus, a thermophilic bacterium. In the collection, since the enzyme is an endoenzyme, preferably the mold is centrifuged, filtered, collected, suspended in a buffer solution, the mold is mechanically crushed by ultrasonic wave, high-pressure homogenizer, French press, etc., solubilized, and purified by column chromatography. |
