RECOMBINANT DNA, ITS PREPARATION, BACILLUS SUBTILIS CONTAINING THE SAME, AND PREPARATION OF PROTEASE USING SAID BACILLUS SUBTILIS

PURPOSE:To increase the yield per unit bacterial cell, and to reduce the amount of contaminant, by preparing a recombinant DNA composed of protease-coding gene integrated in the DNA-introducing vector for Bacillus subtilis. CONSTITUTION:A chromosome DNA containing a protease-coding gene is fragmentized and the obtained DNA fragment is integrated into the DNA-introducing vector for Bacillus substilis to obtain a recombinant DNA. Bacillus subtilis or its affinity strain is transformed with the recombinant DNA, and a strain producing protease out of the bacterial cell is selected from the transformed Bacillus subtilis or its affinity strain. The recombinant DNA, e.g. PAN2 is recovered from the selected Bacillus subtilis or its affinity strain.
