PURPOSE: To provide an electrophoresis method wherein a glycosylated hemoglobin HbAlc can be separated from unglycosylated and other kinds of glycosylated hemoglobins and no interference with abnormal hemoglobin is indicated, and further, a component can be analyzed by concentration measurement after separation. CONSTITUTION: An agarose of 10g is, while agitated vigorously, poured in a distilled water of 830ml. The mixture is gradually heated to 95 deg.C, and then cooled down to 55 deg.C. While agitating, an acid or acids or multiple kinds, and a concentrated solution of 170ml containing a base or bases and a preserving agent are added, and further, chondroitin sulfate and an additive which provides a desired gel property are added. After homogenization, an agarose gel is cast on an appropriate supporting body. An excessive buffering liquid is removed from the obtained gel, and an entire blood or a hemolytic substance of erythrocyte is added according to a general rule of electrophoresis, so that a hemoglobin percentage in a sample is quantified. |
