PURPOSE:To prepare a thermophilic amylase, in high yield, by culturing a novel microbial strain belonging to Escherichia genus and capable of producing thermophilic amylase. CONSTITUTION:A DNA fragment separated from the chromosome DNA of Bacillus thermophilus, having a molecular weight of 2.9X10<3>kbp, coding thermopholic amylase protein, and having a scission site with BamHI in the DNA chain, a scission site with SalI 2.4X10<2>bp downstream of the above site, a scission site with PstI further 0.5X10<2>bp downstream, and a scission site with BamHI further 1.4X10<3>bp downstream, is integrated in a plasmid vector, and is introduced into a host microorganism. The obtained microorganism belonging to Escherichia genus, e.g. Escherichia coli HB101/pHI301 (FERM-P No.7009) is cultured, and the objective thermophilic amylase is collected mainly from the cultured microbial cells. |
