PURPOSE:To produce a considerable amount of a polymeric substance represented by an enzyme protein, by inoculating and culturing a microbial strain belonging to Escherichia genus and containing a plasmid integrated with a specific DNA in a medium containing inorganic salt. CONSTITUTION:A microbial strain belonging to Escherichia genus and containing a plasmid obtained by integrating a DNA carrying a gene information participating in the extracellular selective production of polymeric substance which is a metabolic product of microorganisms of Bacillus genus into a plasmid (vector- DNA) proliferous in the cultured cell, such as colicine E1 factor, etc. which is known as an extrachromosomal gene (plasmid) of Escherichia genus [Eschericia coli HB101 (pEAP2) (FERM-P No.6939)], is inoculated in a conventional medium for the cultivation of microorganisms of Escherichia genus and containing 0.5- 2.0% of an inorganic salt such as NaCl, and the strain is cultured from the latter stage of the logarithmic growth of the microorganism to the stage to finish the production and accumulation of the polymeric substance in the medium (i.e. for 16-48hr) to produce the polymeric substance out of the microbial cells. |
