PURPOSE:To prevent coloration by cellulose in filter paper and to enable measurement of protein in bodily fluid such as urine up to a high concn. by using ''Coomassi Brilliant Blue-G-250'' as a dye, making the high concn. soln. of such dye strongly acidic and impregnating the soln. in the filter paper. CONSTITUTION:1g ''Coomassi Brilliant Blue-G-250'' (hereunder expressed as CBB) is dissolved in 500ml, 0.1% an aq. methyl cellulose soln. 300ml1-2N hydrochloric acid and 150l 1M sodium acetate anhydride are mixed with said soln. and refined water is added to make the entire volume to 1l. Filter paper is impregnated in the soln. and is dried at 50 deg.C. The concn. of CBB is increased and the soln. is made strongly acidic in the above-mentioned way. The intensity of the red purple color that appears when the filter paper is immersed in a sample of bio-bodily fluid such as urine is proportional to the content of protein. The protein in the sample is measured up to a high concn. by utilizing such paper. |
