Collagenase (I) produced by Flavobacterium spec CRZV.1 and having the following characteristics is new:- a) it has a mol . wt. of 83000-105000 daltons; b) an isoelectric pH of 7.9; c) optimal collagenolytic activity at pH 7.6 in 0.05M tris HCl 0.003M CaCl2 buffer at -30deg.C; d) it does not have non-specific proteolytic activity and is weakly inhibited by cysteine and thiol function blocking agents. Used for the tenderisation of meat and in medicine, for the treatment of burns in healing of wounds in dental transplantation, for resorption of discal hernias and the dissociation of cells. Prepn. of (I) comprise fermentation of Flavobacterium spec. CRZV.1 in a suitable medium of 15-35 deg.C. and 30 deg.C. and pH 7-9 pref. 7-6, followed by extraction and purification Suitable media are based on collagem or substances having similar proteic sequences, e.g. polymeric collagem, thermally denatured calhogen (gelatins) or meat extracts, and contain 1-25g/l of material esp. 10g/l of meat extract or 12g/l of collagens. The culture medium also contains vitamins, usually yeast extracts in amounts of 0.1-25g./l, esp. 2.5g/l. |
