PURPOSE:To simply and automatically analyze indoleacetic acid, by a method wherein an indoleacetic acid-containing living body specimen is passed through a pre-treating column under a dessociation suppressed condition to separate impurities therein and to adsorb indoleacetic acid which is, in turn, eluted by a neutral moving phase to be introduced into a high speed liquid chromatography. CONSTITUTION:An indoleacetic acid-containing specimen such as urine or the like is introduced from an introducing apparatus 3 and strong acid type buffer solution with pH of about 2 containing phosphate or citrate capable of suppressing the dissociation of indoleacetic acid is supplied to the apparatus 3 as a pretreating moving phase through a change-over valve 1 and a pump 2 to be mixed with the specimen while the obtained mixture is sent to a pretreating column 6 provided in a thermostatic tank 8 from a flowline change-over valve 5 to adsorb indoleacetic acid by a packing agent for a revers phase coromatography such as a polyvinyl ester type packing agent while impurities in the specimen is removed and discharged. In the next step, a neutral buffer solution with a pH of 7.0 is flowed into a high speed liquid chromatography column 7 from the column 6 as a separation moving phase by changing over the valve 5 and indoleacetic acid separated and eluted in a detection part 9 is subjected to measurement of fluorescent intensity by a fluorescent photometer 9 and the concn. thereof is displayed by a recorder 10. By this method, unstable indoleacetic acid is quantitatively analysed in high preciseness. |
