<p>PURPOSE:To obtain the titled compound, a constitutional unit of lipopolysaccharide existing in the cell wall of Gram-negative bacterium, useful as a drug and agricultural chemical, more easily by synthesis, by reacting a protected saccharide as a starting raw material with a dihalophosphite, etc. CONSTITUTION:A protected saccharide (e.g., 1,0-methyl-2,3,4-tri-O-benzyl-glucopyranose, etc.) shown by the formula R1-OH (R1 is protecting group-containing saccharide residue) is reacted with a dihalophosphite shown by the formula R2OPX2 (X is halogen; R2 is haloalkyl) in a solvent such as preferably THF, etc. in the presence of a base, to give a compound shown by the formula I . This compound is reacted with a protected saccharide shown by the formula R3-OH (R3 is protecting group-containing saccharide residue) to give a compound shown by the formula II, this compound is treated with an oxidizing agent to give a compound shown by the formula III, which is dehaloalkylated and the saccharide-protecting group is eliminated to give the desired diglycosylphospho diester shown by the formula IV (R'1 and R'2 are saccharide residue).</p> |
