A polarographic electrode assembly comprising a confined working electrode is described. The electrode assembly is adapted to fit against a portion of a thin layer chromatogram to form a polarographic cell in which the electrodes of the electrode assembly are all in contact with an isolated zone of the thin layer containing a spot of substance to be assayed. A method of using the polarographic electrode assembly to assay materials separated by thin layer chromatography without elution of the spots containing the substance is also described.
