<p>PURPOSE:Human leukotic T-cells are cultured in a medium containing 8-azaguanine to produce effeciently HGPRT-deficient human leukotic T-cell line. CONSTITUTION:A strain of human leukotic T-cells is cultured in a medium prepared by adding 2muM of 8-azaguanine (8-AG) to the PRMI1640 culture medium containing 10% fetus cow serum (FCS) (according to Flow Laboratories) for a week. Then, its surviving cells are cultured in a medium containing additional 16M 8-AG, further the cultivation is continued as the concentration of 8-AG is doubled until it reaches 100M. Thus, the surviving cells are collected to obtain the objective human leukotic T-cells deficient in hypoxanthine-guanine-phosphoryribosyl transferase (HGRT). 6-Thioguanine or 6-mercaptopurine rivoside may used instead of 6-thioguanine.</p> |
