In the method according to the invention, the sample to be investigated is suspended in alkali hydroxide, and concentrated hydrazine hydrate, a first auxiliary agent and a second auxiliary agent for the purpose of colouring are added to the suspension. The sample is shaken and heated at 80 to 95 DEG C, preferably 90 DEG C for 5 to 30 min. A mixture of dilute hydrochloric acid, ammonium chloride, an organic solvent mixture and solutions of Phloxin B and Fast Green FCF are then added to the sample. The sample is coloured, cooled and filtered through a peripherally hydrophobized cellulose nitrate filter. The filter is washed with a solution containing a surfactant and hydrochloric acid and having a pH of 3.5 to 4.5 and dried in a drying oven. The dry filter is laid with the coated side on a glass support and transferred with a clear liquid to a transparent cellulose nitrate film.
