PURPOSE:To prevent the clogging of a color developing reaction cell or the like while improving preservative stability of a reagent, by carrying out photometry of an amino acid mixture in the reagent after separate amino acid solution eluted from a separation column is mixed with a spesified ninhydrin reagent composition at a specified rate. CONSTITUTION:Individual amino acids are separated from a sample containing an amino acid mixture such as body fluid with an eluent from an ion excharge chromatography column and the obtained amino acid solutions are mixed with a ninhydrin reagent in a mixer section. The composition of the reagent is as follows: 1.8-2.2%(W/V%) ninhydrin, 41.2-48.8% dimethyl sulfoxide, 33.8-26.2% of methyl cellosolve, 0.9-1.1M of lithum acetate, 0.1-1.2M of acetic acid, and 0.08-0.15% of hydrindanethione. The mixing ratio of the amino acid solution and the ninhydrin reagent is set at 1:1-2 by volume. This enables continuous photometry of individual amino acids in a short period of time without clogging due to precipitates generated in a thermal color developing reaction cell or the like. The reagent can be preserved stably for as much as 1-2 months in an atmosphere of N2 gas. |
