PURPOSE:To measure the concentration of amicacin in blood quickly and to prescribe a medicine safely, by combining amicacin labelled with glucose-6- phosphoric acid dehydrogenase with anti-amicacin antibody conflictingly with amicacin and measuring enzyme activity after reaction. CONSTITUTION:Labelled amicacin is obtained by reacting amicacin with glucose- 6-phosphoric acid d hydrogenase for safety medication of remedy amicacin for bacteria infections symptoms. On one hand, amicacin is immunized to a rabbit and an antibody is obtained from its blood and then, a calibration curve based on a competitive reaction of the mixture of the marking amicacin and already known quantity of amicacin for the antibody, is found preliminarily. The concentration of amicacin in blood of a patient is determined by said calibration curve. In the case where the specimen is blood plasma, citric acid or EDTA is used for an anticoagulant. Measurement is obstructed by using heparin. Thus, amicacin is determined in high sensitivity and good reproducibility. |
