<p>Detrm. of the capacity of cells (or cell fractions) for supporting efficient transcription of a specific mRNA corrsp. to a specific protein is effected by (a) contacting a nucleic-acid-free extract of the cells with an exogenous DNA sequence contg. a gene coding for the same protein, in the presence of necessary precursors (including nucleotides) in vitro, and (b) detecting if any of the mRNA is formed. Conversely, exogenous DNA sequences can be tested for mRNA transcription by contact with a nucleic-acid-free extract of cells known to support transcription. The method can be used for early diagnosis of diseases associated with the prodn. or non-prodn. of specific proteins, e.g. anaemia, leukaemia or viral infections.</p> |
