PURPOSE:To measure population of five kinds of lymphocytes easily and simultaneously, by letting lymphocytes in peripheral blood separated from bright red blood of a subject react with a minute plastic particle covered with anti-human immunoglobulin and a sheep red blood cell. CONSTITUTION:A lymphocyte is separated from haparinized peripheral blood collected from an empty stomach in early morning of a subject. To this lymphocyte dispersion are added a suspension of an immunobead 3 obtained by covering a minute plastic particle with antihuman immunoglobulin in a Hanks' solution and a suspension of a sheep red blood cell that has absorbed an inactivated fetal calf serum. A rosette of the lymphocyte formed by the reaction among the three sepcies above is observed with a microscope on a slide glass coated with a specific pigment. Five kinds of lymphocyte, i.e., a B cell 4 to which only the bead 3 easily recognizable because it has been dyed has adhered, a T cell 1 to which only the red blood cell has adhered, a D cell to which the bead 3 and the red blood cell 2 have adhered together, a phagocyte 6 eaten voraciously by the bead 3 and a naked null cell are measured separately. Thus with one reaction five kinds of specimen are measured simultaneously for a short period with a little blood-gathering. |
