Interferences encountered in the use of hemolyzed samples in the process of quantitatively determining the amount of iron in a serum sample are minimized by maintaining the incubation of the sample in a buffer system for an extended period, e.g. at least about 5 minutes. The compound 9-(2-pyridyl)-acenaphtho¢1,2-e!-as-triazine or its sulfonated derivative may be used as both a chelator and indicator of ferrous ion.
