<p>Prepn. of immunoglobulin for intravenous injection comprises treating blood plasma or fractions obtd. by subjecting blood plasma to cold alcohol fractionation, with acid; and then fractionating the resulting prod. using alkylene oxide polymer or copolymer with mol. wt. 2,000-20,000. Acid treatment is by keeping the starting material under acidic conditions at 3-10 (4-10) deg.C for 30-180 mins. The pH is 3.2-5.0, pref. 3.8-4.2, and the ionic strength is 0.002-0.30, pref. 0.1-0.20. The protein concn. at this time is pref. 2-10 w/v.%. The acid used is inorganic acid such as hydrochloric acid or organic acid such as acetic acid. Alkylene of the alkylene oxide polymer is e.g. methylene, ethylene, propylene or butylene. Fractionation is conducted at pH 4.6-5.4, pref. 4.8-5.2. The obtd. supernatant has added to it 10-15%, polyethylene glycol and the pH is adjusted to 7-8 to ppte. the immunoglobulin. This can be recovered by centrifuging. The method gives immunoglobulin in high yield.</p> |
