<p>PURPOSE:To make possible to isolate a plant protoplast having excellent activity by treating plant cell with an enzyme solution in the presence of an adsorbent such as an active carbon adsorbing volatile component or phenols. CONSTITUTION:An adsorbent such as an active carbon or a gel for hydrophobic chlomatography, etc., capable of adsorbing a volatile component or phenols is dispersed in an agar, solidified and pulverized. Then, the pulverized agar is dispersed in an enzyme solution prepared by adding an osmosis pressure controlling agent such as mantol, etc., to an enzyme such as cellulase or pectinase, etc. Next, textures of stem, leaf or root, etc., of a plant of Perilla or Pelargonium, are finely cut and added to an enzyme solution, then treated, for instance, at about 25-30 deg.C for 2-6 hour. By said process, a substance making an activity of protoplast depress, ejected from damaged protoplast, is effectively adsorbed and removed, thus plant protoplast is isolated without depression of activity of protoplast.</p> |
