<p>A process for testing chemical and biological substances (suitable for pharmaceuticals, foodstuffs, insecticides, pesticides, solvents and other goods in daily use) for harmful effects, and for eliminating these side-effects was described in the parent patent. The substance to be tested is administered to experimental animals and after sufficient time the number of plasmapolyp labelled cellorganell-poor protrusions in the blood plasma or in histological slices from tissue is determined and by positive precipitation, the substance being tested is combined with a cpd. which reduces the formation of plasmapolyps. The plasma polyp number in the blood plasma is determined by; (a) the heavy blood cells are sepd. by partitioning the sample with a soln. of a copolymer of sucrose and epichlorohydrin (m.wt. ca 70000) and centrifuging; (b) sepg. the plasma polyps in the sample freed from heavy blood cells by diluting with physiological salin centrifuging and sepg. the sediment; (c) diluting this sediment with Krebs-Ringer soln. centrifuging and sepg. to further enrich the sediment, so that the centrifuge rate increase from step (a) to step (c); and (d) counting the plasma polyps in the sediment from (c) by resuspending and observing in a phase contrast microscrope. The determination of plasma polyps is simplified and the time necessary is reduced. The margin of error in step (d) is below 0.19.</p> |
