PURPOSE:To prepare an antibiotic substance, in high productivity, by contacting immobilized cells of antibiotic-producing bacteria with a nutrient medium containing a quite limited amount of carbon source and a nitrogen source consisting exclusively of an organic nitrogen source. CONSTITUTION:Bacteria capable of producing an antibiotic substance, e.g. Bacillus polymyxa ATCC 10401, etc., are immobilized, e.g. by polyacrylamide gel process, etc. The immobilized bacteria are made contact with a nutrient medium containing only organic nitrogen sources such as peptone, bouillon, oatmeal, etc., in a concentration of 2-0.1 W/V% to accumulate a large amount of the antibiotic substance in the medium. Continuous preparation of a high-titer antibiotic substance is also possible by the continuous contact reaction introducing the nutrient medium and oxygen continuously into the reactor. |
