Preparing cpds. of general formula(I) AcOCH2CH2CH2CO2R (I) Ac = acyl R = an ester residue, e.g. alkyl Analgesics. I (Ac = CH3CO, R=Et) is twice as effective as acetylsalicylic acid and has LD50 (p.o., mouse) 1900 mg./kg. (a) KOAc + XCH2CH2CH2CO2R (I) X = hal (b) I + R'OH I (R=R') + ROH If R'OH has higher b.p. than ROH Ethyl 4-chlorobutyrate (60 pts.), PhCH2CO2K (107 pts.), KI (4.6 pts.), and Me2SO (15 vol.), were heated 24 hrs. at 150 deg., cooled, dissolved in H2O (500 pts.), and extd. with Et2O (2X200 pts.). The Et2O was dried, evapd. and the residue distilled, giving I, b.p.135 deg./0.5 mm. |
