<p>Somatic cell count in milk is measured by means of fire-fly luciferin-luciferase assay of adenosine triphosphate (ATP) in the sample. The milk sample is treated with a non-ionic surfactant to release ATP. The method detects mastitis in cows, quickly, accurately, and economically and is suitable both for field testing at farms and in laboratories. Suitably the concn. of ATP is measured in a luminescent photometer after addition of a firefly luciferin-luciferase reagent in a buffer having a pH value between 74-80 and a Mg content of 0.005-0.02 moles/l. The ATP concn. is converted to the number of somatic cells per ml. by applying conversion factor determined by measurement of a known standard.</p> |
