PURPOSE:In isolating and purifying a physiologically active substance produced by a gene recombinant, to obtain the physiologically active substance having both high degree of purification and removal ratio of pyrogen, by heat-treating an aqueous solution of the physiologically active substance in a coarsely purified state or in an advanced purified state. CONSTITUTION:In purifying a physiologically active substance that is produced by a gene recombinant, having cell disorder activities against L-M cell and properties to cause hemorrhagic necrosis reaction at a tumor site when it is administered to BALB/c mouse to which Meth A sarcoma cancerous cell is transplanted, at any stage purification process, preferably in a crude stock solution or after coarse purification of one stage, an aqueous solution containing the physiologically active substance is heat-treated preferably at 6-8pH at 50-65 deg.C, especially at 58-62 deg.C for 5-180min, especially for 30-60min. By this heat treatment, the nucleic acid, lipid, and protein derived from molds are made into insoluble precipitates, degree of purification is extremely improved by the succeeding gel filtration, and removal of pyrogen is simultaneously made easy. |
