<p>PURPOSE:To stick an attachment cell to a culture carrier by simple operation, to extend the cell and to efficiently culture the cell by treating a culture mixture of attachment cell with protease in an aqueous medium to release the attachment cell, treating the attachment cell with an immobilized protease inhibitor and culturing the attachment cell. CONSTITUTION:An attachment cell [e.g. human kidney cancer derived cell TRC-29R strain (FERM-P 10274)] is cultured together with a microcarrier in a medium, made into a culture mixture of cell wherein the attachment cell grows thick on the microcarrier, the culture mixture is centrifuged to separate a supernatant liquid, which is suspended in an aqueous medium such as buffer solution and treated with a protease (e.g. trypsin) to release the attachment cell from the microcarrier. Then the attachment cell is cultured while adding an immobilized protease inhibitor (e.g. p-aminobenzamidine-agarose beads) so that the attachment cell are stuck to the microcarrier, extended and efficiently cultured.</p> |
