PURPOSE: To evaluate the level of theophylline in sample by bringing the sample into contact with alkaline phosphatase and specific phosphoric acid ester, and comparing the current generated by the electrochemical oxidation of electroactive compound with the current due to the standard electrochemical oxidation. CONSTITUTION: The testing element has an operating electrode and collating electrode. The operating electrode is separately carried to avoid the mutual action of alkaline phosphatase and electrically inactive phosphoric acid ester. When sample is added, liquid is brought as substrate into contact with enzyme in addition to theophylline existing in the sample, left to stand for 60 to 150sec at constant temperature to be reacted, then the current held and generated at the operating electrode at 150mV and monitored for 30 to 60sec, and read. The quantity of the theophylline can be obtained by using the current by collating it with the current generated under the standard conditions by using the theophylline of known concentration. |
