Prodn. of a compsn. for preventing viral haemorrhagic septicaemia (VHS) of trout comprises first culturing the virus on fish cells, then inactivating it totally with beta-propiolactone (I). The virus used is pref. cultured on the EPC (epithelium papulosum cyprini) cell line to a titre of 104-109, esp. 5x107 to 5x108, plaque- forming units per ml. After deactivation with (I) the safety of the prepn. is pref. tested by 2 cultures on fish cell lines, and then opt. further treated with aq. HCHO to a final HCHO concn. of 0.05%. The vaccine protects trout again VHS more conveniently than the rigorous disinfection procedures normally used, and long-lasting protection is afforded.
|
