Pure ergosine and its acid addn. salts are prepd. from residual mixts. of alkaloids, obtd. is by-prods. in the prepn. of ergotoxine alkaloids, by chromatographing on alumina and elution with a mixt. of 1,2-dichloroethane and butanol. The ergosine is recovered from the eluant and opt. converted to salts. Ergosine and its salts are used to treat migraine. The methane sulphonate has LD50 i.p. in mice of 162mg/kg and i.p. in rats of about 400mg/kg. Pharmacological tests (effect on arterial blood press., effect on heart, antiadrenaline effect, effect on tonus of bronchi, antiseratonin activity) show that ergosine is a catecholamine antagonist on the alpha-adrenergic receptors, a serotonin antagonist on the D-receptors, improves smooth muscle tonus and has a positive inotropic effect on the heart. It is administered enterally or parenterally in daily doses of 1-70 mg/kg. |
