Optical resolution of aryl-pyridyl-alkyl tert. amines - by reaction with optically active maleic acid

Prepn. of optical isomers of arylpyridyl alkyl-substd. tert. amines of formula (I): (where R is dialkylamino, morpholino, piperidino, pyrrolidino or imidazolino X is H or halogen; n=1-4) comprises reacting racemic (I) with L- or D-malic acid. The resulting L- or D-malate is then purified by repeated recrystn. from various solvents, and the crystallised malate is converted into L- or D- (I) by treating with alkali metal hydroxide in a medium comprising H2O and a water-immiscible organic solvent. The other isomer is recovered from the recrystn. mother liquors. The d isomers of cpds. (I) are antihistamines, and the 1 isomers are local anaesthetics. Optically active malic acid is less expensive than the acids proposed in FR1258226.
