PURPOSE:To attain excellent separation of three components, an aspartic acid, a glutamic acid and glutamine, by executing analysis while the flow velocity of a buffer solution sent to a column for separation of an amino acid is changed in stages or a linear gradient manner. CONSTITUTION:Out of buffer solutions 1 to 4 and a column regeneration solution 5, one buffer solution is selected by a series 6 of solenoid operated valves. By changing over the degree of opening of the series 6 of solenoid operated valves, these buffer solutions 1 to 4 are sent for separation at a slow flow velocity at the time of start, and then the flow velocity is increased for executing analysis in the latter half of a process. A buffer solution pump 7 sends the selected buffer solution to a separation column 10 via ammonia filter column 8 and an autosampler 9. An amino acid separated in the column 10 is mixed by a mixer 13 with a ninhyrdin reagent 11 sent by a ninhydrin pump 12 and reacts therewith in a reaction coil 14. The amino acid produced in the reaction is detected continuously by a photometer 15 and outputted as a chromatogram by a data processing device 16. |
