PURPOSE:To obtain the subject new enzyme gene coding an ultra-heat resistant beta-galactosidase derived from Pyrococcus furiosus having a resistance to a surface active agent and capable of industrially producing an enzyme used in fields of a food industry and saccharide engineering, etc. CONSTITUTION:This new enzyme gene for producing an ultra-heat resistant beta-galactosidase codes an ultra-heat resistant beta-galactosidase having an amino acid sequence containing an amino acid sequence expressed by the formula or a part of the amino acid sequence, being thermophilic, excellent in heat resistance, derived from Pyrococcus furiosus having a resistance to a surface active agent such as SDS, etc., and is used for production, etc., of ultra-heat resistant beta-galactosidase in fields of food industry, saccharide industry, etc. The gene is obtained by culturing Pyrococcus furiosus DSM3638, collecting the bacterium, bacteriolyzing the bacterial cell, recovering a genome DNA according to an ordinary method, preparing the library and screening the library. Furthermore, a plasmid obtained by integrating the gene is introduced into a host cell and manifested to provide the objective ultra-heat-resistant beta- galactosidase. |
