PURPOSE:To efficiently extract an active ingredient in cells from a mold containing chitosan in a cell wall, by using chitosanase, a cell wall dissolving enzyme. CONSTITUTION:A mold belonging to the order Mucorales or Moriliales is cultivated in an agar medium to form a spore, which is transplanted to a liquid medium and the mold is multiplied to accumulate an active ingredient in the cell. The prepared cell is directly dispersed in an aqueous solution or a buffer solution or washed and dispersed, mixed with a cell wall dissolving enzyme containing chitosanase derived from Bacillus pumilus BN-262 (FERM P-8814) and slowly shaken at 30-40 deg.C for 6-8hr to extract the active ingredient. beta-1,3- Glucanase, cellulase or chitinase may be used as the cell wall dissolving enzyme except chitosanase. |
