<p>One of the objects is to realize the complete denudation of amniotic epithelium by a simple operation with as little damage to the compact layer and basal membrane as possible. Another object is to provide a method capable of treating a number of amnions simultaneously. A method for preparing an epithelium-denuded amnion comprising the following steps of (1) preparing an amnion isolated from a living body; (2) subjecting the amnion to a freezing and thawing treatment; (3) treating the amnion subjected to the freezing and thawing treatment with trypsin; and (4) washing the amnion subjected to the trypsin treatment is provided.</p>
