PURPOSE:To perform the quantitative analysis of the specific antigen or antibody in a specimen, by immobilizing the antibody or antigen on a liposome having a marker substance sealed therein and blocking the immobilizing functional group remaining on the surface of the liposome. CONSTITUTION:An antibody or antigen is immobilized on a liposome composed of one of phospholipid and glycolipid and having a marker substance sealed therein according to a crosslinking method to constitute a reagent. After the antibody or antigen is immobilized, the immobilizing functional group remaining on the surface of the liposome is blocked through the reaction with an SH- group-containing substance or an amino group-containing substance. As a result, the functional group extincts and no non-specific reaction is generated between the protein and trace chemical substance in a specimen. Since there is correlation between the outflow quantity of the marker substance due to the destruction of the liposome and the quantity of the substance to be examined in the specimen, the quantitative analysis of the specific antigen or antibody in the specimen can be performed by measuring the outflow quantity of the marker substance.
