The invention relates to a method for identifying protein biomarkers directl y in a specific region of interest of a tissue which comprises the identification o f a mass of interest by analysis of MALDI imaging mass spectrometry results, followed by the identification of the protein represented by said mass of interest by direct elution from the specific region of interest of a tissue, fractionation and tandem mass spectrometry.
