METHOD FOR PRETREATMENT OF TISSUE MATERIAL OF MAMMAL

PROBLEM TO BE SOLVED: To provide a method for the pretreatment of a mammalian tissue material for the preparation of a specimen for PrPSc (scrapie-type prion protein) detection and measurement to complete the pretreatment with reduced number of steps in a short time without exerting adverse effect on the detection and measured value of PrPSc. SOLUTION: The pretreatment method contains a solubilized PrPC decomposition step to add a decomposition enzyme mixture containing a surfactant and proteinase K to the collected mammalian tissue material and homogenize the product. The invention further provides a step to add a DNA decomposition enzyme and/or an RNA decomposition enzyme and/or a collagen decomposition enzyme to the decomposition enzyme mixture.
