Oligonucleotide (I), for use in primer extension reactions, comprising a 5'-target-binding sequence (A), a non-target-binding sequence (B) and a 3'-target-binding sequence (C), is new. Independent claims are also included for the following: (1) a pair of (I) as amplification primers; (2) a kit comprising a pair of (a) and one or more detectors able to detect an amplicon; (3) detecting absence or presence of a target nucleic acid (tNA) in a sample using a pair of (I); (4) amplifying a target sequence in tNA using a pair of (I); (5) generating single-stranded DNA in a primer-extension reaction using three (I); and (6) a DNA molecule (Ia) that is any of 42 specified oligonucleotides, the fully defined sequences of which are given in the specification.
