A method for detecting the presence or absence of a target molecule suspected to be present in a sample, comprising: a) contacting the sample with a polymer scaffold bound to a fusion molecule, wherein the fusion molecule comprises a target molecule binding domain configured to selectively bind to the target molecule, and wherein the fusion molecule is non-specifically bound to the polymer scaffold; b) loading the polymer scaffold into a device comprising a pore that separates an interior space of the device into two volumes, and configuring the device to pass the polymer scaffold through the pore from one volume to the other volume, wherein the device comprises a sensor configured to identify objects passing through the pore; and c) determining, with the sensor, whether the target molecule binding domain is bound to a target molecule upon translocation through the pore, thereby detecting the presence or absence of the target molecule in the sample.
