PURPOSE:To obtain the titled substance useful as an anticoagulant of heparin, a drug for sustaining medicinal effects of insulin, an enzyme stabilizer and an antibiotic stabilizer in high purity and in high yield, by extracting milt of fish containing protamine with a mixed solution of a mineral acid and a salt thereof. CONSTITUTION:Milt of fish (e.g., herring, salmon, trout, mackerel, etc.) containing protamine is extracted with a mixed solution of a mineral acid 9e.g., preferably 3-20wt% sulfuric acid, hydrochloric acid, etc.) and a salt thereof (preferably ammonium sulfate or sodium sulfate). Then, the extracted solution is cooled so that a protamine mineral acid salt is precipitated and recovered. The concentrations of the acid and the salt of the supernatant liquid are adjusted and reused as the solution for extraction.
