The purification of long-chain acyl-coenzyme-A synthetase which involves solubilizing the enzyme with a surfactant and subjecting the solubilized enzyme to affinity chromatography and the enzyme, acyl-CoA synthetase, purified thereby. The enzyme is useful in the preparation of a labelled acyl-CoA, and in the assay of free fatty acids in blood.
