PROBLEM TO BE SOLVED: To simply produce the subject compound which is useful in the treatment of cancers, rheumatoid arthritis, diabetes complication, central nervous system disorder by culturing a proper microorganism, for example, Oomycetes, collecting its mycelia and isolating the objective sphingolipid from the mycelia. SOLUTION: A microorganism in Oomycetes, for example, Pythium ultimum (ATCC 26083) is inoculated into a culture medium, subjected to the shaking culture at 25 deg.C for 48 hours. Then, the mycelia are collected by filtration on a glass fiber filter, rinsed with water and freeze-dried. The cells of the freeze- dried mycelia are charged in screw tubes containing chloroform, methanol and water, they are homogenized by means of a homogenizer for 30 seconds, then chloroform and water are added to the homogenized product to cause a phase separation. The lower chloroform phase is recovered and subjected to a semi-preparative HPL chromatography whereby the objective sphingolipid represented by the formula [R<4a> is C(CO)R<8a> (R<8a> is a 13-23C alkyl, a 13-23C (poly)alkenyl) is simply obtained. |
