The present invention provides modified aminoacyl-tRNA synthetase (ARS) which has improved reactivity with an N-methyl-amino acid compared with naturally occurring aminoacyl-tRNA synthetase. The modified aminoacyl-tRNA synthetase according to the present invention can make it possible to aminoacylate an N-methyl-substituted amino acid, e.g., N-methyl-phenylalanine, N-methyl-valine, N-methyl-serine, N-methyl-threonine, N-methyl-tryptophan and N-methyl-leucine, into corresponding tRNA with higher efficiency compared with naturally occurring aminoacyl-tRNA synthetase. According to the present invention, it is possible to produce a polypeptide containing an N-methyl-amino acid with higher efficiency. |
