发明名称 APPARATUS FOR AUTOMATICALLY DETERMINING ARRANGEMENT OF NUCLEIC ACID BASE
摘要 PURPOSE:To continuously detect fragments of nucleic acid which are separated by migration by such an arrangement wherein an electric field is applied in the direction perpendicular to the direction of migratory separation at such a position slightly higher than the positive polar terminal of an electrophoresis supporting body. CONSTITUTION:As a nucleic acid specimen identified by a radioactive isotope is placed on the side of negative pole of a gel 2 and both ends of the gel are caused to contact with an electrode solution 1 and the specimen is caused to migrate by a voltage 9 of approximately 50V/cm (length of gel), ingredients having the same molecular weight form migratory bands 4 respectively and migrate from the negative pole to the positive pole at such a speed which is substantially the inverse proportion of the logarithm of their molecular weight. On one hand, buffer solution for rendering migratory band which is the same in composition as the solution of the gel is caused to slowly flow through a hole 11 of a glass plate 3 provided at a position slightly nearer to the negative pole than the positive pole end and at the same time, a voltage 10 of such a degree that will not greatly disturb potential slope in the direction of migration is applied to the solution. By this, separated migratory bands 4 pass through a radioactivity detector 6 and the quantity of radioactive ray is measured and they are recovered and stored in a waste recovery tank 8.
