FACTOR IX CONFORMATION-SPECIFIC MONOCLONAL ANTIBODY

PURPOSE:To obtain a monoclonal antibody having specificity exclusively to factor IX having a conformation varied with Ca<++>, by separating an antibody- producing cell from an animal immunized with coagulation factor IX and cloning the obtained cell. CONSTITUTION:A BALB/C mouse is immunized twice with factor IX purified from fresh frozen plasma and with FCA adjuvant supplied by DIFCO Co., and the spleen cell is extracted from the immunized mouse. The cell is fused with a mouse myeloma cell (P3-X63-Ag8-U1) and cloned to obtain 8 kinds of hybridoma (No.1-No.8) capable of producing a monoclonal antibody having specificity to factor IX. Each hybridoma is proliferated in the abdominal cavity of a mouse. Among the monoclonal antibodies produced by the above process, the antibodies of clone No.4, 5, 6 and 8 can specifically recognize only the factor IX conformation caused by Ca<++> and does not recognize conformations caused by the other metals.
