PURPOSE:To obtain protein.kinase derived from a procaryote, having independence to cyclic nucleotide and excellent stability, capable of phosphorylating rice globulin and swine plasma protein, by cultivating a specific bacterium capable of producing protein.kinase, gathering a bacterium cell from the culture mixture and collecting a membrane fraction. CONSTITUTION:A bacterium such as Micrococcus luteus FK1001 strain (IAM11007), belonging to the genus Micrococcus, having optimum temperature of approximately 65 deg.C at pH7$.0 for 3min reacting time and independence to cyclic AMP or cyclic GMP, cyclic nucleotides, capable of phosphorylating rice globulin and swine plasma protein and of producing protein.kinase is cultivated in a medium consisting of glucose, ammonium sulfate, yeast essence and an inorganic salt at pH6.8-7.8. Then the culture mixture is centrifuged to gather a bacterium cell. The collected membrane fraction is incorporated with a buffer containing Triton X-100, stirred, subjected to solid-liquid separation, the supernant liquid is obtained as a Triton X-100 soluble membrane fraction and used as the above-mentioned protein.kinase. |
