Provided are a vibrio universal suicide vector for gene knockout, a construction method thereof and a use thereof in the vibrio gene knockout. The suicide vector pLP12 is a circular vector comprising a P BAD promoter, a repressor protein gene araC, an RP4 transfer initiation site, a chloramphenicol resistance gene, an R6K replication initiation site, a multiple cloning site region, and a lethal gene vmi480, wherein the multiple cloning site region comprises at least two AhdI enzyme cleavage sites. The suicide vector pLP12 is digested with AhdI to form a linearized suicide vector pLP12T.
